Action Spectroscopy¶
Record a light-triggered product or response across excitation wavelengths instead of measuring only how much light a specimen absorbs.
Core Idea¶
Action spectroscopy varies excitation wavelength and measures an event or response induced by the light—such as ion fragmentation or photosynthetic oxygen evolution—instead of measuring only beam attenuation. An action spectrum is response versus wavelength, not automatically an absorption spectrum. IUPAC distinguishes response per incident photon from an efficiency spectrum per absorbed photon.[ref-f5ca2c0b23c8][ref-154f8b99ff3f]
Scope of Application¶
Gas-phase ion studies can detect photoinduced fragment ions even when the selected ion population is too small for a useful transmission measurement. Bush and colleagues used infrared photodissociation to study cationized arginine. In another setting, Tamburic and colleagues measured oxygen evolution and chlorophyll fluorescence of Nannochloropsis oculata at fourteen visible wavelengths under reported equivalent photon flux.[ref-d7418c3757d4][ref-c4e46f83aa56]
Clarity¶
The action channel must be named. Fragment yield, fluorescence and oxygen production measure different consequences of absorption. Their curves can differ from true absorbance because light may be absorbed without producing the monitored action, and action efficiency may depend on wavelength, fluence and thresholds. “High sensitivity” is a possible advantage, not a defining guarantee.[ref-d7418c3757d4][ref-154f8b99ff3f]
Manages Complexity¶
The method makes a difficult optical event visible through a countable consequence. But it moves complexity into response calibration: unequal photon dose, saturation, sample energy and branching into uncounted products can distort action-band heights. A peak is interpretable only together with the chosen detector and normalization.[ref-f5ca2c0b23c8][ref-d7418c3757d4]
Abstract Reasoning¶
Specify specimen and photoinduced action, scan wavelength under controlled conditions, detect response, and normalize by the stated photon denominator. If the question is functional effect, the action curve can answer it directly. If the question is optical absorption, test whether the action probability is known or sufficiently stable before treating the response as a proxy.[^ref-f5ca2c0b23c8]
Knowledge Transfer¶
Ion fragmentation and algal oxygen evolution share wavelength-selected illumination and response measurement but not response physics. A gas-phase dissociation threshold cannot be imported into a photosynthetic culture. The common method is a wavelength-to-action map, not one universal absorption spectrum or sensitivity claim.
