Internal Standard

A quantitative calibration method that adds a fixed known amount of a distinct reference to every sample and calibrator and uses analyte-to-reference response ratios to cancel shared variation.

Core Idea

An internal standard travels with the analyte through sample preparation and measurement. Because each specimen receives the same known reference amount, variation that changes both responses proportionally can cancel in their ratio.

Success depends on timing and similarity without interference. The standard should be absent initially, added before the nuisance steps of interest, recovered like the analyte, measured separately, and connected to concentration through a validated ratio calibration.

Clarity

State analyte and standard identity, purity and amount, addition stage, matrix absence, extraction behavior, resolution, response definition, calibration equation/weighting, range, blanks, uncertainty, and failure diagnostics. Inclusion test: Require a chemically distinct reference added in known comparable amount to every standard and unknown before the variation it should correct, separate responses, and a validated ratio-based calibration. Exclusion test: Exclude standard addition of analyte, external calibration with no added reference, a naturally present peak used without known amount, and a surrogate that responds differently enough to invalidate correction. Nearest boundary: An isotopically labeled analogue is an internal standard when separately measured; standard-addition calibration instead spikes varying analyte amounts into the sample. Exit condition: The method fails when reference is endogenous or interfered, added too late, not separately resolved, or affected differently from analyte across samples. Common misclassifications: It is not standard addition of analyte. It is not external calibration alone. Any convenient peak is not automatically a valid standard. Ratios do not correct analyte-specific loss. Nearest named distinctions: External standard: Is not added to every unknown. Standard addition: Spikes the analyte at varying levels. Calibration check: Verifies performance but may not accompany each sample. Recovery surrogate: May monitor processing without satisfying quantitative internal-standard assumptions.
